Phenobarbital (or phenobarbitone) is a long-acting barbiturate medication primarily used as an anticonvulsant to treat various types of epilepsy, including tonic-clonic and partial seizures, as well as to manage neonatal seizures and emergency sedation. It acts by increasing inhibitory GABA neurotransmission in the brain. Common side effects include drowsiness, sedation, ataxia, and dizziness. Uses and Indications Epilepsy: Used to control the frequency and severity of seizures, often used in developing countries. Sedation: Acts as a sedative and hypnotic. Withdrawal: Used to treat withdrawal symptoms from other barbiturate dependencies. Mechanism of Action Phenobarbital functions as a nervous system depressant. It enhances the inhibitory effects of GABA (gamma-aminobutyric acid) at the receptor, reducing neuronal excitability. Side Effects Common: Sedation, drowsiness, listlessness, irritability, dizziness, and cognitive or memory impairment. Serious: Respiratory depression (slowed breathing), severe skin rashes (Stevens-Johnson syndrome), liver dysfunction, and increased risk of depression or suicidal thoughts. Tolerance: Sedation often decreases over the first 2-3 weeks of treatment
